Neem in Dermatology: Designing Novel Compounds to Enhance Psoriasis Treatment

Psoriasis is a chronic skin condition that affects millions of Canadians. Although medications exist to treat psoriasis, many cause serious side effects, including liver damage, low immunity, and birth defects. Neem is a plant native to many tropical regions and is recognized for its anti-inflammatory properties. The goal of this research is to optimize psoriasis treatments using neem. In this project, computer simulations were performed to study neem compounds and assess their binding to Tumor Necrosis Factor alpha (TNF-α). Results indicate that the compounds campesterol and β-sitosterol were most stable within the TNF-α binding pocket. However, improvements were necessary to limit side effects. Structural modifications were applied to top candidates and results demonstrated limited hepatotoxicity, carcinogenicity, and immunotoxicity — common side effects associated with current medications. This research marks a significant step toward developing sustainable alternatives which serve as safe treatments for psoriasis.
